New pathway regulates immune balance and offers promising drug …

November 9th, 2010

A new Post “New pathway regulates immune balance and offers promising drug development target” was written on the September 21, 2010 at 9:00 pm on “IC Marketplace”.

ScienceDaily (Sep. 21, 2010) — St. Jude Children’s Research Hospitalscientists have identified a new pathway that helps control the immunebalance through reciprocal regulation of specialized T lymphocytes, which play very different inflammatory roles.

Investigators also determined that two drugs working in different waysto dampen the inflammatory response in patients with multiplesclerosis or following organ transplantation target this newmechanism. Further research into the pathway might lead to newmedications to block other autoimmune disorders or to newanti-rejection drugs, researchers said. The work is published in thecurrent online issue of Nature Immunology.

T cells are the white blood cells responsible for both driving andmodulating the immune response. This work focuses on a mechanism atwork as T cells differentiate into the more specialized T-helper 1(Th1) cells that drive inflammation or the regulatory T cells thatwork to shut it down and protect healthy tissue from a misguidedimmune attack.

“The success or failure of the immune response requires T cells tomake the right decision about their fate,” said Hongbo Chi, Ph.D.,assistant member of the St. Jude Department of Immunology and thepaper’s senior author.

“In this paper we describe the receptor that controls the cell fatedetermination of different subsets of T cells; controlling the choiceto become either an inflammatory or a regulatory T cell,” Chi said.Earlier work from Chi and others linked the receptor, S1P1, to otheraspects of T cell functioning.

Researchers also found surprising evidence T cell response isregulated by a lipid the T cell secretes rather than a protein knownas a cytokine. If confirmed, Chi said the finding would mark the firsttime a lipid, rather than a protein, served such a signaling functionin T cells.

For this study, investigators used both cultured cells and speciallybred mice to link the S1P1 receptor to the fate of the two sub-groupsof T cells. Stimulating S1P1 activates a pathway that drives the cellto become a pro-inflammatory Th1 cell. Th1 cells rally other immunecomponents to act against infection and other threats. At the sametime, S1P1 activation down regulates differentiation of regulatory Tcells. The S1P1-dependent effect on both sub-groups of T cells relieson its ability to dampen signaling through another pathway in the Tcell; this second pathway uses a different molecular route toinfluence the T cell’s fate, working through cytokine TGF-betaactivation of a signaling molecule called Smad3.

“There is a reciprocal change between the two cell subsets. With thissystem, T cells that do not become regulatory T cells have a tendencyto become T helper type 1 cells,” Chi said.

The S1P1 pathway is also targeted by the anti-rejection drugrapamycin, which is used to protect organ transplant patients, andFTY720, which has the promise to become the first oral therapy for useagainst relapsing multiple sclerosis. This study is the first to showboth work in part by modulating this molecular pathway.

The work expands on earlier research from Chi’s laboratory showing theS1P1 receptor played a central role in inhibiting the development andfunction of regulatory T cells. “In this paper, we show that thereceptor controls differentiation of conventional T cells as well,” hesaid, specifically Th-1 cells. S1P1 also plays a role in the movementof T cells throughout the body.

Researchers are now focused on understanding the pathways in moredetail. The questions include how S1P1 activates the mTOR pathway.Once activated, mTOR, a protein kinase complex, works to dampensignaling along the TGF-Beta — Smad3 pathway, thereby promoting Th1cell differentiation at the expense of regulatory T cells.

The paper’s co-first authors are Kai Yang of St. Jude and GuangweiLiu, formerly of St. Jude and currently of the Chinese Academy ofSciences. The other authors are Sharad Shrestha of St. Jude and SamirBurns, formerly of St. Jude.

This work was supported in part by the National Institutes of Health, the Arthritis Foundation, the Lupus Research Institute and ALSAC.

wmbb.com – Autistic Child's Battery Charges Stir Controversy

October 1st, 2010

An 11-year old boy is facing felony battery charges after an outburst at Merritt Brown Middle. However, his family believes the school should receive some blame for poor handling of his autistic background.

“We get more calls because they can’t handle it. They say they can but then when it comes down to it, are you really getting an education? Is there really enough for these teachers to deal with these kids?” said the boy’s aunt, Jayanna Green.

The boy is 1 of an estimated 4,600 students in this district placed in an Exceptional Student Education Program. The school board disagrees with his family arguing that the program is up to par, particularly for students with autism.

“It’s probably been the exceptionality not only here but nationwide, it’s grown the fastest within the last decade,” said ESE Director, Pat Martin.

Martin believes the district does a good job given the circumstances. He argues there’s a plan designed by parents and teachers to help every disabled child.

“We do have students who basically have assistance, individual assistance, throughout the school day,” said Martin.

According to the family, that may be what set off the boy the school sent off in handcuffs.

“He gets aggravated that the helper walks beside him, he’s one of the few kids at school with a helper, he gets picked on for that,” said Green.

Martin understands, saying there must be a balance.

“There’s kind of a fine line there that they have to give them some time to step away to be a student or a teenager, to communicate with their friends,” said Martin.

At this point, the family has chosen to home school the child.

DOJ Enthusiastically Welcomes First Lady

June 23rd, 2010

Good morning. Thank you all for being here today to help me welcome our nation’s First Lady – and my good friend – to the Department of Justice.

In the recent past, including many miles on the campaign trail, I’ve had the pleasure of getting to know our First Lady. And I can tell you that, like the President, she has a brilliant legal mind. I have been so impressed by her legal skills that I’m going to make her an offer- right now- to join the best lawyers in the world, right here at DOJ.

I’ve also learned that she has a deep appreciation for the work and many responsibilities of the Justice Department. But what’s impressed me most, and what I admire most about her, is her commitment to justice.

Many of you already know her extraordinary story – that she grew up in a small apartment on the South Side of Chicago and, with hard work, determination and the support of a loving family, made her way to Princeton University, then Harvard Law School, then on to one of the nation’s premier law firms. And she decided long ago – long before she became First Lady – that she wanted to harness the power of the law to generate positive social change and build a more just society.

That commitment took her in unexpected directions. As she once put it – and I know many of you feel this way, too – she realized that she, “wanted to have a career motivated by passion and not just money.” And so she built on her legal training to serve communities, assemble volunteers, and – despite the pay cut – spend her time inspiring young people to enter public service themselves.

And did I mention that it was because of the law that she met a certain summer associate – and her law firm mentee – who would change her life? She has said that she, and I quote, “wasn’t expecting much” of the young Harvard Law student who everyone else was raving about. But shortly after they met, our President summoned all the charm he could muster – and all the moves he had – and apparently it worked. From that time on, our First Lady has been, not only a distinguished attorney, executive, and community leader, but also, in her husband’s words, “the rock” of her family. Indeed, she does seem to do it all: lawyer, advocate, visionary and, above all, the mother of two wonderful daughters, a supportive and engaged wife, and a wonderful daughter herself.

Over the past year and a half, the First Lady has also become “the rock” of our nation – a committed, and already accomplished, force for positive change, especially for young people. Last month, I had the privilege to join her in Detroit, where she kicked off a day of mentoring and called on young students to work hard and, just as important, to give back. And her “Let’s Move!” campaign to eliminate childhood obesity is already creating a healthier – and, in a very real sense, more just – America.

But her commitment and her tireless efforts don’t stop there. She also works to support military families, to serve as a role model for working women, to promote the arts and arts education, and – of course – to continue to make sure that my boss still takes out the trash. That can’t still be true!

When I think about the First Lady, I’m struck by the fact that, though I’ve only known her for a few years, it feels like so many. That’s the kind of friend our speaker is. From the day we met, she has made me feel welcome and at home. And so in that same spirit, I’d like us to welcome her to our home here at the Department of Justice. Ladies and gentlemen – the First Lady of the United States, Michelle Obama.

MRS. OBAMA: Thank you. Thank you so much. (Applause.) Thank you, everybody. Thank you. Such a warm and wonderful welcome. I am thrilled to be here.

I want to start by thanking our outstanding Attorney General, Eric Holder, your boss, for that very kind introduction, and also for the wonderful work that he’s doing here at the Department of Justice. He is — I could say the same accolades as he said about me. He’s just been a phenomenal support, not just to the President but to me personally.

As he mentioned, he joined me along with celebrities and other people from the administration in Detroit to do some very important mentoring in Detroit. And he was just amazing. I mean, you know how busy he is. And my view is that if this man can take the time out to fly and spend a day talking to young people, I mean, sitting down at a table with kids, and talking about how they can pursue their dreams, how he can use his own story to show them that they can reach for passions that maybe they thought they never could, that he, in his own role, serves as a role model. If he can do that, then we all can do that.

And I know that there’s so many of you here who are following that lead. And I’m grateful to him and I’m grateful to all of you for serving in that role. So we have to give him an incredible thank you. (Applause.)

I’m told that Eric started out as a 25-year-old law graduate — school graduate working in the Public Integrity Section here at DOJ. You were 25?

ATTORNEY GENERAL HOLDER: That was five years ago.

MRS. OBAMA: Five years ago. (Laughter.) And even though he’s been around the block a few times since then — (laughter) — only five years — he’s never lost that sense of responsibility that comes from working to uphold our highest legal principles. It’s a responsibility that all of you share, and one that some of you have been shouldering for quite a while, I understand. That’s why I want to take a moment to recognize the folks here on the stage with me. These are some of the longest-serving employees here at the Department of Justice. I don’t know the numbers here, but they’ve been here for quite some time, and I want to take some time to give them a round of applause for their dedication. (Applause.)

It’s just wonderful to see people who have made commitments for decades to government service, and it’s important for the world to see, particularly young people, to see how people are building and have built lifetimes here serving the broader community.

And I know that even though we’re here at Main Justice, I also want to recognize the men and women who serve as the faces of this agency in communities all across the country: the FBI and the ATF agents. (Applause.) The U.S. Marshals and the hardworking folks at the U.S. Attorneys offices who are on the ground every day — yay, yes — (applause) — they’re keeping us safe and protecting our most basic rights.

And when I travel, one of the great things I get to do is usually see the U.S. Attorneys on the ground. So our congratulations and thanks goes out to everyone.

One of the privileges of being First Lady has been the opportunity to visit so many agencies over the past year or so so that I can thank all of you, really, for the hard work and dedication that you’ve all put in. You put in long hours. And a lot of people look at the President, they look at your boss, and they say, well, you’re working hard. But the truth is — and we all know this — you all are putting in that kind of time as well. You’re making sacrifices. You miss time with your families. And often, you do it without getting any recognition from anyone.

So I want to let you know how much that we value everything that you’re doing here, however long you’ve been doing it, because I know we have a lot of newbies here, folks who are just joining the department as well. Yay, all right, let’s give them a round of applause, too. (Applause.)

So that’s one of the reasons I’ve been doing these visits, to make sure that you all know that even in the heat of change and all the work that goes on here, that we haven’t forgotten the work that you do and the sacrifices that you make.

These visits, though, also help me get a better understanding of what’s happening in some of these agencies, to listen, to learn about your work and to help spotlight the difference that you make in the lives of so many Americans, because when I show up, there are cameras that usually come, and I think it’s important for the people around the country to know that government is working hard for the American people and that it’s made up of everyday Americans who are making sacrifices on their behalf.

And I have to admit that I’m especially excited to be here at DOJ because we have a lot in common, many of us here. As many of you know, long before I lived in the White House, I worked in Chicago, and I did a little law thing. (Laughter.) I decided to study law for some of the same reasons many of you did. Number one, math was really hard. (Laughter.) And as my mother said, I talked a lot — (laughter) — and could write pretty good. But it’s also because I’ve seen the power that law has to change people’s lives in a very real and meaningful way. And I knew that lawyers had the ability to help turn words on a page into justice in the world –- to keep a neighborhood safe; to keep a family in their home; to leave our children a world that is a little more equal and a little more just.

And I also — as Eric mentioned — I met this guy named Barack Obama while I was studying law. (Laughter.) Yes, he was my mentee — a summer associate when I was a first-year associate. So that was a nice little perk from my law career. (Laughter.)

And here at DOJ, you all represent the ideals that drew us all to this business in the first place: those principles of equality, fairness and the rule of law. Your responsibility is not to a particular party — and that’s important for people to understand — or to a particular administration or to a President. You work for the American people. You do battle every day on behalf of the most vulnerable among us. And you touch the lives of virtually every American in ways large and small -– even if they don’t realize it.

For a department that started out with a single, part-time employee in 1789, the workload here at DOJ has really never stopped growing. And I know you all are feeling that right now.

Whether it’s keeping our nation safe from terrorist attacks, or bringing our most hardened criminals to justice, protecting consumers or safeguarding our civil rights, your work has never been more important that it is today.

That’s especially true in the wake of the worst environmental disaster that we’ve ever faced here in this nation. And I know that the Attorney General and several members of the leadership team have traveled to the Gulf, and many folks here in this agency are working tirelessly to ensure that accountability is going on, that we’re protecting taxpayer dollars, and that we’re helping those affected by the oil spill really get back on their feet.

And people need to know that the Department of Justice is at the center of that work. But it’s not just the work that you do that makes this place so special. It’s what you all bring to the work that you do. It’s the passion, and the persistence and the energy that you bring to your cases.

And I know to be here, taking pay cuts as many of you do, you’ve got to be doing it because of passion because all of you all would be at a firm somewhere if it didn’t mean something to you.

But that’s true whether you’re an attorney, a paralegal, a librarian, a support staffer — truly, the dedication that you’ve all shown is extraordinary. And I’m proud — very proud — of the work that you’ve done, and I’m extremely grateful for what you’re doing every day.

And it is not an easy job. That I know as well. But the fact that so many of you have stuck around for so long really says something about the culture of this agency.

Administrations, as you know, can come and go, but the pride that you put into your work, it never fades. As Attorney General Holder likes to say, working here isn’t just about making a living. And that’s so important for young people out there to know and to see. These jobs, it’s not about earning the dollar; it’s about making a difference in someone’s life.

And this group really takes those words to heart. I’m told that in the first six months of this year, your attorneys have taken on 20 pro bono cases -– from custody battles and landlord-tenant disputes, to domestic violence and personal injury cases. Pro bono, for those of you who don’t know, is completely free legal service.

And 50 of your attorneys, I understand, have staffed legal clinics right here in D.C., helping to write wills, to file taxes and to do other important work for members right here in this community who couldn’t otherwise afford it.

In the end, that’s really what the Department of Justice is all about. That’s really what the field of law is supposed to be about. You all help make the promise of our laws a reality for every single American regardless of their race, their standing or their political affiliation.

From the Great Hall of the Supreme Court to a folding table in a legal clinic, you help our families secure the protection that they need and the rights that they deserve. And you do it with a level of fairness and compassion that stands as an example to us all.

So for that reason, I’m here to show you, along with the rest of America, our gratitude, our admiration. These are going to be tough times. And we’re going to need every one of you to buckle up and work even harder. But it’s easier to have that conversation here because you all know what hard work means. You all know what sacrifice means.

And it’s important for us to share those values with the next generation. We need to replace you all. We need to start working on the next generation of staffers and attorneys and librarians and paralegals who are going to fill these seats in decades to come. And they’re going to do that because of the work that they see you doing. They’re going to do that because of the pride that you take in your work. We are the role models for the next generation.

So we are grateful for your work. And I just look forward to coming out there and shaking a few hands.

So thank you, thank you so much. (Applause.)

John Devilman's Tattoo Grue

April 24th, 2010

Tattoos are a form of expression that express individuality, but also demonstrate their wearers’ predilections and tastes, allowing them to bond in much the way horror enthusiasts do. “Seeing what can be done with tattoos these days, thanks to them being accepted into the mainstream, people are getting more creative with their desired pieces,” skin artist John Devilman says.

The melding of tattoos and horror is a fairly recent phenomenon that has sparked a creative revolution in both communities. “Horror tattoos are always popular,” Devilman says, “because they can be sexy, nostalgic, intimidating and fun. And even as tattoos become more and more mainstream, they’re still a bit more rebellious. They can also be a kind of litmus test as to who fellow fans of your favorite cult movie may be.”

There has been a resurgence of both fright fare and tattoos in the last decade that has led both to become more commonplace and accepted in “normal” culture. Conventions around the country (including Fango’s own) are frequented by fright fans riddled with tattoos. Both passions are now perceived as lifestyles, and the inked images represent a way to connect the body and favorite films together. “I know many horror fans who’ve used tattoos to help express their affinity for such films,” Devilman notes. “It speaks more clearly and more faithfully than any Pinhead shirt or a trowel signed by Kyra Schon might.”

Devilman’s Zombie Tattoo shop in West Hollywood is helping bring together this form of art and the affection for all things scary, and the artist also tatted Fango fans and even staff, including this writer, at our Los Angeles convention last year (pictured here). “We thought it would be a great chance to promote our shop,” he says of the event. “I mean, where better than FANGORIA to represent Zombie Tattoo? We knew that most of the people who would attend would be just like us and relate to our work and our shop’s direction. It was such a fun convention, and we felt right at home with all the other horror fans there.”

Like many, Devilman found his way to Los Angeles with hopes of landing an acting gig, but it was his artistic talents that garnered him a secure foothold. “My sworn brother came from Shreveport, LA and told me to come live with him,” Devilman recalls. “He said that he had talked to this guy who owned a tattoo shop out there about me and my artwork, and that guy would be willing to take me in and show me how to tattoo.” Having nothing to lose, Devilman took the chance, figuring tattooing “would be better than flipping burgers. So three weeks and a speedy, makeshift apprenticeship later, I was tattooing.”

A string of odd occurrences brought the establishment into existence—fitting for a shop called Zombie Tattoo. As the legend goes, the place’s two owners thought up possible names and threw them into a hat. The discovery seemed pretty unique, and it was fortuitous that they found themselves in West Hollywood, of all places. “Some psychic to the stars suggested to the owners’ wives that they find a place here—that it would bring them great fortune or some such mystic stuff,” Devilman says.

For his tattoos, Devilman draws inspiration from many different avenues, implementing aspects from various artists into the detailing of his work. “My line quality, I’m sure I get from Vaughn Bodé of Cheech Wizard fame,” Devilman notes. “When I was younger, I was in love with his comics work. My insane detail was a bit of my own until, after seeing my work, a friend suggested I take a look at Geofrey Darrow [HARD BOILED, BIG GUY, RUSTY THE BOY ROBOT]. I took some influence from him in high school and have since adapted it to suit my personal style.

“As far as art that I like, I’m currently falling back in love with old monster magazine art—Basil Gogos’ [of Famous Monsters of Filmland fame] stuff especially,” he continues. “I also like some of the ‘Stories of Real Men’-type magazine art that has commandos door-kicking some Nazi compound as they’re about to set upon a buxom, young, near-naked beauty with a ridiculous instrument or arcane method of torture. As for comics I draw inspiration from, CONAN has had a pretty good run, with the art being a blend of sketch and fine painting. I also enjoy the art Jae Lee does on the DARK TOWER: THE GUNSLINGER series. Storywise, 100 BULLETS, FABLES, Y THE LAST MAN, EX MACHINA, CONAN, INVINCIBLE and THE WALKING DEAD are the titles I currently pick up.”

It takes a certain kind of person to work in a horror tattoo shop, and Devilman is definitely up to the challenge of holding down the position, as he’s more than happy to share his affection with like-minded customers. “I grew up on horror movies—the good, the bad and the ugly,” he says. “Given the name of our shop, I’ve had the opportunity to do tattoos that come from the minds of fellow genre fans. They’re always way more fun than putting a tramp stamp of some soon-to-be-ex-boyfriend or tribal armband on someone”—and more original, for that matter.

Devilman proves his horror-fan mettle when he names his favorite films, which include titles from all over the spectrum. These include the first and third NIGHTMARE ON ELM STREET films, EVIL DEAD II and ARMY OF DARKNESS, along with SHUAN OF THE DEAD, PUMPKINHEAD, ALIENS and John Carpenter’s THE THING. Devilman also has a multitude of further ideas for tattoos that seem to draw from everywhere and anywhere. “I want to really do a BLADE RUNNER piece, some Frank Frazetta stuff, Dr. Herbert West, Vincent Price, Sloth from THE GOONIES, Al Bundy, Burt Reynolds, Freddy, another Beetlejuice, Predator…shoot, the list goes on and on, but you get the idea.”

As for the basic question of why horror is so appealing to him, he offers a very recognizable, honest answer: “I think monsters are amazing. I still remember when I was in second grade, all the kids in my class would have me draw their monsters fighting Freddy or Jason or something. I always hung around older kids because I was too young to watch those types of movies, but they would have them and we’d watch GHOULIES or TROLL or something, and I’d get to see all this gore and chicks with their boobs showing running and screaming from a really rad bogeyman. What about that wouldn’t appeal to a youth?”

In addition to the horror tattoos the Zombie shop is known for, Devilman also creates a diverse array of other artwork. “I do a lot of portraits, horror or otherwise,” he says. “I’ve had a string of people coming in asking me to draw them pinup girls from scratch and make them look realistic; that’s always a challenge.” Some of his favorites among these have included a Gogos-esque rendition of Frankenstein’s Monster, a Barbarella back piece and Ash from EVIL DEAD II.

Speaking of portraits, Devilman has one other recurring, and especially personal, theme he has recently begun. “I’m going to start tattooing myself on as many people as possible,” he reveals. “I’ve already got another tattoo artist to have my face inked on them, and a few others to agree to get my lovely mug on them somewhere as well. I also tattooed my phone number on an old boss, and I’ve promised him that I won’t ever change it because of that.”

Even though tattooing is a demanding job, Devilman always has fun with it, in part due to his customers. “One client is a really good guy who enjoys joke tattoos,” he explains, “and he had me conjure up a tattoo with a bunch of sad dinosaurs in the style of Don Bluth’s LAND BEFORE TIME congregating around while a meteor crashes down above them with a banner that says, ‘God Doesn’t Believe in Us.’ ” Devilman even runs a special promotion, “where I give free tattoos away on April 1. The catch is, it’s got to be a design I want to give. The person can decide not to get it, but then I just go down the line until I find someone who’ll want it. From this, I did my ‘chickenf**ker’ tattoo and another one that was a cute rubber ducky with a banner over it stating, ‘I watch you masturbate.’ Good times.”

Zombie Tattoo itself is riddled with accoutrements that any horror aficionado can appreciate. “Our shop’s got a lot of zombie toys, movie props, posters, etc. Our financial outfits worn by zombies from SHAUN OF THE DEAD, RESIDENT EVIL movies, some of the gas masks and replica guns—it helps the whole horror vibe of the shop. I love it.”

And for anyone reading this who is thinking about getting a tattoo, Devilman has a piece of advice for you. “Sometimes, people just come in wanting anything. They haven’t really thought it through, and I’ll usually try to convince them to put a little thought into what they’re getting, or else they’ll end up with something they’ll regret.” Once someone has chosen a piece that they want for sure, Devilman is enthused to oblige. “If it’s something that I really want to do, that’s when I want to tattoo them. Or if they tip well!”

There are a huge number of kid’s toys available in the market, but it is important that you choose the right toys for your kid that will help boost his/her imagination and thinking abilities while they are not even aware of it. There are many such toys including character toys like those of Sylvanian Families, Shrek, Hannah Montana, Star Wars, Disney Fairies and cartoons; and also board games which are very good for improving the aptitude and logical abilities of kids. In addition to this there are many outdoor games as well, which help in improving the physical potency of the kids.

Being a parent it is your duty to decide on what type of toys you should buy your child to mould him accordingly. For all the little kids, Sylvanian Families are a very popular option. They initially originated from North America and Japan in the 1980s, when they became very popular.

Sylvanian Families are actually anthropomorphic animal characters which were made much alike humans, who were supposed to talk, walk and live their lives with their families as human beings do. The Sylvanian Family characters touched the imagination of the kids to a great extent and thus, many kids started collecting their own Sylvanian characters in order to make a full family and house-hold for them.

The toy-makers this made a guinea pig family that included characters Andrew, Peter, Penny, Nicola Pettyfur and also their own guinea pig babies Simon Pettyfur and Lindsay Pettyfur. The Pettyfur family was supposed to stay together, eat together and sleep together in the same house just like humans do. Thus, the toy makers also started making creating toy houses which had little beds, coffee table, dining tables, sofa sets and chairs, to give it a more realistic look.

The coffee tables also came with tiny cups and plates, where the Sylvanian family was supposed to have their morning and evening tea and snacks. Kids just love pretending to be part of such a family, because they control everything in the family according to their imagination. So what are you waiting for? Buy your child the Sylvanian Family collection and see his/her imagination sore.
