Stem Cells Take Cues From Fluid In The Brain

Brain cells in the cortex — the part of the brain responsible for cognition, learning and memory — multiply and move to their appropriate position between the second and third trimester of embryonic development in humans. but until now, researchers have had little luck finding the molecular signals that direct the process as well as determining how the signals get delivered to the cells that need them.

The current team extracted cerebrospinal fluid from mouse embryos around two weeks after conception, when their brains develop most quickly. the fluid contained high levels of a protein, insulin-like growth factor or Igf2, which is known to help stem cells multiply and differentiate. Notably, the protein isn’t elevated after birth. when the authors blocked Igf2, stem cells in the brain stopped making brain cells, which resulted in abnormally tiny mice brains. And when the team placed brain stem cells in a dish filled with Igf2-rich, embryonic cerebrospinal fluid, the cells proliferated rapidly. "this was clearly the environment the stem cells needed to be happy," LaMantia explains.

Brain cell proliferation is only a good thing when the time is right, however. After all, unrestrained cell multiplication leads to tumors. according to this report, Igf2 knows it’s time to activate in the fluid because of proteins in long cells that surround the fluid. ‘Hall Pass’ disappoints with ridiculous plot, characters

Farrelly brothers’ comedy devoid of laughs, lacks any semblance of entertainment value

A foolproof way to tell that a film is in the lowest tier of comedies is if, during the previews, the trailer for the latest Tyler Perry disaster (which he calls a movie) contains more laughs than the film you paid to see. the billing of the Farrelly brothers’ latest bastardization of a film, “Hall Pass,” as comedy is the worst entertainment fraud since Milli Vanilli.

One could argue these judgments are a tad extreme; however, I think they actually sell the film short. In fact, I am saddened I do not possess a versatile enough vocabulary to fully encompass the abhorrence that gurgled deep in the pit of my stomach while Owen Wilson’s whiny and obnoxious voice grated my eardrums for two-hours.

The Farrelly brothers’ signature films rely on extreme humor (usually crude gags), and while the movies may be pretty awful at times, they aren’t usually boring. “Hall Pass,” however, manages to achieve that mediocrity while being excruciatingly hum-drum, which truly is testament to the brothers’ devolution as filmmakers.

The film centers around the concept of a week free from the duties of marriage. Granted this freedom by their wives, the main characters — two middle-aged, married men — can do literally whatever they want. the theory behind this inane concept is the age-old adage, “Absence makes the heart grow fonder.”

If the hall-pass concept seems like a disastrous relationship tool, it’s probably because it is delivered by Joy Behar, who plays Dr. Lucy, a voice of reason. Naturally, instead of just sleeping with the foxy neighbor or secretary, Rick (Owen Wilson) and Fred (Jason Sudeikis) explore their freedom through lame shenanigans.

For the first hour of “Hall Pass,” the theater was completely silent, without one giggle or chuckle. Toward the end of the film, it became obvious the Farrelly brothers took an additional 30 minutes just to add completely idiotic and ridiculous plot points, presumably because the law of averages states at least one of them has to be funny.

“Hall Pass” paints each of its characters as gender stereotypes, with the men characterized as insatiable sex fiends who only have one thing on their minds and the women as uptight and sexually aversive. It is like watching CBS sitcoms for two hours. Stereotypical characters, however, were not the downfall of this film though.

In fact, the Achilles’ heel of “Hall Pass” was that it was completely devoid of all traces of humor.

If you are a fan of the Farrelly brothers’ 2007 work “The Heartbreak Kid,” then go see “Hall Pass.” but if you are dying to see it, just go ask your parents to detail their sex life meticulously for you. It should pretty accurately replicate the experience of sitting through “Hall Pass.”

What’s relatively surprising about this film—and the franchise in general—is just how consistently creative it remains considering its abundant reinterpretations. For a franchise that has had numerous interpretations of its plot throughout the course of nearly 30 years, Toki wo Kakeru Shojo is still able to offer yet another unique take on the time traveling heroine. While the device of time travel has always been a main component within telling the story, the films have steadily remained focused upon the personal dilemmas of its characters, never allowing the use of time travel to take over the course of its story. This is even more visible within this film, mainly because the element of time travel is only performed twice throughout and doesn’t remain a huge aspect of the film like some of its predecessors. at its core, the film focuses upon the notion of promise and how one can fulfill that promise despite the passing of time. This notion was explored in the original 1983 film, and it works exceptionally well here even more so given the rather vivid relationships shared amongst the characters. While the familial bond was only a minor aspect of the 2006 animated film, here it’s utilized as the catalyst to promote the story and is viewed as major element in concluding the storyline from the 1983 film.

With this film focusing primarily on the familial relationships shared amongst its cast, it once again explores the platonic love that develops between the main heroine and an individual from the past. This aspect, while certainly a considerable element of the film’s latter half, is not entirely realized here. While one can see their relationship as authentic in many regards—and the emotional backing is expressed adequately as well—the way it develops is treated rather hastily. This is rather unfortunate considering that the film provides great background concerning the maternal relationship shared between Akari and her mother, but does little in showcasing the development of Akari and Ryota’s relationship, which becomes an important element as the film nears its conclusion. what we are offered is a relationship that is seemingly presented as one-sided throughout the film, but is eventually expressed as a mutual one only when the direst of consequences present themselves. While the emotional impact is still prevalent, more time spent exploring the fruition of their relationship would’ve made the conclusion considerably stronger and in a better sense, simply more believable.

Overall, Toki wo Kakeru Shojo is still a wonderful addition to the established universe of Yasutaka’s novel, even if its somewhat inadequate in terms of fully developing its relationships. Considering the longevity of the series, the aspect of originality is certainly brought into question, but the element of creativity is still present. This quality stems primarily from its characters, especially that of actress Riisa Naka who does an effective job at revitalizing and representing the role of the heroine of the series. Fans of 2006’s animated film will appreciate her honest and quirky portrayal within this film, which establishes a new approach as to how one may view the main heroine in the Toki wo Kakeru Shojo series. and while the usage of time travel is not as eccentric as past films, the film’s ample focus on familial affairs and long withstanding promises remains at the forefront. whether this is the final film in the series is certainly up for speculation, but what Toki wo Kakeru Shojo provides is a satisfying and complementary conclusion that is sure to please fans—both new and old alike—of its classic tale of time travel, love, and loss.

Local Fox affiliate fattens up on news

when Jerry Martin took over as general manager of WXIN-TV Channel 59 in 2004, the Fox affiliate was airing a three-hour local news and entertainment show on weekday mornings and a half-hour news show at 10 every night. Eighteen and a half hours of news a week. This January, after Channel 59 expands its weekend morning news shows by an hour, the station will be airing 54-1/2 hours of live local news each week. And if everything goes as planned, by the end of 2012 it will add a 6-7 p.m. newscast weekdays, bringing the total to an astonishing 59-1/2 hours. That means more than a third of its airtime will be devoted to local news—about twice as much as its three competitors and maybe more than any other network affiliate in the country. “We’re really trying to change the way people look at Fox59,” Martin said. “we want them to think of us primarily as a news station with Fox prime-time programming. I think that will happen because of the number of hours we produce. And as the marketplace changes, we believe that the one with the most resources devoted to news, the one that’s on the most, is going to be seen as the premier news place in the market.”

If nothing else, it’s certainly generating gasps in the industry. “Wow—59 [hours],” marveled mark Kraham, chairman of the Radio Television Digital News Association and news director of a station in Hagerstown, Md., that does 29 hours of local news each week. “That’s an incredible amount.” as Martin sees it, Channel 59 is doing what it has to do to be able to control its own destiny. for decades, stations like his typically existed on a heavy diet of syndicated programming—daytime talk, court shows, game shows and sitcom reruns. But now there’s less of that programming available, what is available can be costly and, because of cable, little of it is available exclusively. News, on the other hand, is local, exclusive content that can also be used to feed the station’s website and shared through mobile devices like smartphones and iPads. “really, it’s local television going back to its local roots, more so than just running whatever Hollywood produces,” he said. And it appears to make sense in other areas. Martin said the more news Channel 59 does, the higher its web numbers have been. In the past year, he said, website traffic has quadrupled, and the site is making money for the station. “not the kind of money you make running a TV station, but it’s starting to become a significant part of our business plan,” he said. Local news also serves as a profit center for affiliates. not as much as prime time—where programming historically has been free and the profit margin from the local advertising time sold during prime time is nearly 100 percent—but as much or more than syndicated programming. “If you negotiate a good price [for syndicated shows], you’re going to be looking at fairly good margins—nearly as good as you would have in news,” Martin said. he wouldn’t specify a dollar value for either. Savvy strategy? despite all this, opinions differ about whether this strategy will work in the long run. John Cardenas, general manager of WTHR-TV Channel 13, Indianapolis’ highest-rated local news station, acknowledged that if “you want to grow your news image, it’s probably not a bad strategy on their part.” But Cardenas remained skeptical that Channel 59 could provide enough fresh content to retain viewers. or that viewers would gravitate to Martin’s station. WXIN, which in September 2009 launched the only local news show from 4-5 p.m. weekdays, initially had lower ratings in that hour than when it aired the syndicated talk show “Maury.” the newscast now fares better, running in third place. so far, Channel 59’s launch of a 5 p.m. newscast this September hasn’t yet translated into a ratings surge. It’s in fifth place—even behind reruns of “Everybody Loves Raymond” on sister station WTTV-TV Channel 4, Cardenas said. “If you look around the country, a lot of stations have been expanding their newscasts in part to meet the changing lifestyles of viewers, but I think a lot of times it’s strictly to get advertiser revenue the station isn’t getting,” Cardenas said. “My sense is that’s probably the case here, given all the different times periods they continue to expand their newscasts in.” Martin counters that he expects the 5 p.m. newscast to finish October with a 1.9 rating/4.3-percent share of the audience, a 25-percent improvement over a year ago when “are You Smarter Than a 5th Grader?” aired in that time slot. And since WXIN sells advertisers on its demos—the demographic breakdown of its audience—not total number of household viewers, he’s happy with what he’s seen so far. “I would say it is doing exactly what we thought it would do at this early stage … and it will probably be no. 3 in time period in demos by February,” he said. Since Martin launched his strategy, the station’s news ratings overall have improved, too, especially in the mornings, though it generally trails Channel 13 and WISH-TV Channel 8 during time periods when the stations compete head to head. And Martin points out that the idea isn’t to get people to sit down and watch full three- or four-hour blocks of news each day but to give them a place to stop for 15 minutes to catch up on what’s happening in the city and state. If they know Channel 59 is likely to be airing news, they’ll be more likely to turn to the station and, Martin hopes, bookmark fox59.com and follow the station on Facebook and Twitter. ‘Local, local, local’ RTDNA chairman Kraham said the organization doesn’t track the number of news hours stations air, but adding more local news is definitely a nationwide trend. His station just added a 7 p.m. newscast as a way to reach commuters getting home later. “And the thing that I keep hearing from everyone is that, to remain relevant, to compete with all of the other voices and things that are out there calling for attention—everything from social media to online products—you must do local,” he said. “make sure you remain local, local, local, providing a unique product that is unavailable anywhere else.” things used to be different. Mike Ahern got his start in Indianapolis TV in 1967, when local news consisted of a half hour a night. In 2004, when he retired from WISH as the market’s gold-standard anchor, the station had been calling itself “24-hour News 8” for more than a decade and was airing around 30 hours of news a week. Ahern hadn’t heard of Channel 59’s plans to expand, but “my first thought is, that sounds like an awful lot of makeup,” he said, laughing. then he got serious. “I’ve always thought if anything survives this multi-channel, multi-informational age we’re in, it would be local news.” is there that much news to cover in Indianapolis? Ahern doesn’t think so. “If you hang in there for the full hour and a half or two hours of news, it becomes terribly repetitious. I think they’re counting on, in a very mobile society with lots of choices out there for news sources, people just grabbing their news on the run. In that case, it’s probably not a bad idea.”•

Rio Tinto’s $116bn iron ore venture with BHP Billiton is close to collapsing after regulators objected to the deal, the Times writes.

Rio was forced to issue a statement last night saying that no formal decision had been made on the future of the project. Nevertheless, both companies are understood to believe that European regulators’ disapproval makes it unlikely that the deal can succeed in its present form. The proposed joint venture would have seen Rio and BHP integrate their vast iron ore operations in the Pilbara region of Western Australia, the Times adds.

Warren Buffett, the billionaire investor, has hit out at pay practices on Wall Street, attacking the lack of reform despite two years passing since the financial crisis struck. “People have a propensity to gamble, and it gets made easier and easier for them,” Mr Buffett told a conference in Washington DC yesterday. “One of the problems we still have is we have unbalanced incentives for managers of huge financial institutions.” In future, chief executives of banks who need government assistance should “go broke”, said Mr Buffett. Their wives “should go broke, too”, he added, the Telegraph reports.

Kazakhmys stoked controversy over its links to the dictatorial Kazakh government yesterday when the company’s chairman and biggest shareholder Vladimir Kim sold an 11% stake in the miner to the Astana administration.
